Abstract
Free fatty acids (FFA) are toxic to cells, and thus sequestered in the form of triacylglycerides (TAG) in cytoplasmic organelles called lipid droplets (LD) that bud from the endoplasmic reticulum (ER). We previously reported that ER-lysosome tether Mdm1 spatially regulates LD biogenesis at ER-lysosome contact sites to maintain ER lipid homeostasis. Deletion of Mdm1 disrupts this process and leads to FA-induced lipotoxicity. Our recent finding reveal that deletion of ER-lysosome tether Mdm1 also dysregulates uptake and metabolism of essential amino acid methionine. This suggests a previously unrecognized role for ER-lysosome tethering in regulating the crosstalk between FA and methionine metabolism. In this seminar, I will present supporting data for our central hypothesis that Mdm1 functions in coordinating ER-localized lipid metabolism with methionine sensing at the lysosome and discuss current directions for my lab.
